Countercurrent chromatography
Countercurrent Chromatography is one kind of liquid-liquid chromatography which doesn’t need solid bearing material or under prop. It is regarded as a useful tool in the separation and preparation for natural medicine, for it can not only avoid bearing material absorption and pollution to samples, but also has a high retention volume which is adapted for large input volume preparation. But for a long time there is little research in the CCC system integration and automation which make against this technology’s application and popularization. This research aim to make a fundamental model for designing safe, auto and convenient High Speed Countercurrent Chromatography (HSCCC) system that utility the one-direction Hydraulic Dynamic System Equivalent (HDSE), take practice on the IV-Type Coil Planet Centrifuge field by the PTFE coil tube. Below is the research work: 1) Design Centrifuge. Including drive method, coil tube brace design, coil tube brace fixation, axletree fixation, etc. Determine the important parameters for semi-preparation HSCCC such as centrifuge brace length LB , brace flange diameter DB, centrifuge flange diameter DC, etc. Discuss the instrument manufacture material, radiator and β alteration method. 2) Use computer to adopt the spectrophotometer signal, give continuous absorption in real-time. 3) Design a new and handy sample fraction collector, give the control module code. 4) Use computer to manager HSCCC system. By taking AD/DA interface technology and writing program, the system can measure HSCCC rotation speed,display the absorption curve in real-time, precisely and automatically judge absorption peak and collect the fraction. 5) Installation and performance evaluation. Use this HSCCC system to separate and prepare Daidzin and Genistin soybean insoflavones, discuss pump impulse influence to baseline, as well as mobile phase flow, centrifuge rotary speed and βinfluence to retention phase volume. Validate this HSCCC system’s separation capability by the good separation result in optimism condition.
